Candida albicans 64545 cells were stained with SYTO 13 and the agarose matrix was counterstained with streptavidin-R-phycoerythrin conjugate. Red and green fluorescence collected separately using a cooled CCD camera were merged using image processing software. The left panel shows an encapsulated yeast cell immediately after encapsulation. Yeast cells fluoresced green while the GMDs fluoresced red. After incubating for 6 hr, the yeast cells formed colonies within the GMDs (right panel). The average mean fluorescence for encapsulated microcolonies is higher than the fluorescence from single encapsulated cells.
