Fluorescent Profile of a Sorted Population Enriched for High Secretors
Approximately 104 GMDs which exhibited PE fluorescence intensity in the highest 7% of the population were sorted. The gel matrix was digested with agarase and the recovered cells were cultured for three weeks. At this point, both the sorted population and the original cell line were again encapsulated and analyzed for secretion. The PE fluorescence histogram for GMDs containing cells from the original, unsorted cell line is shown in Panel A and the enriched cell line histogram is shown in Panel B. The data shows that the average secretion from the enriched population was approximately fivefold higher than that of the original cell line (mean fluorescence of 255 vs. 46 units).
